How to Reconstitute Research Peptides (Lab Guide)

Research peptides are supplied as a lyophilised (freeze-dried) powder for stability during storage and transport. Before they can be used in solution, they must be reconstituted β€” dissolved in a suitable sterile diluent. Done carefully, reconstitution is simple; done carelessly, it can degrade the peptide or introduce contamination that ruins an experiment.

What you need

  • The lyophilised peptide vial, brought to room temperature (cold glass encourages condensation)
  • Bacteriostatic water β€” sterile water containing a small amount of benzyl alcohol as a preservative, which allows the reconstituted solution to be stored for longer than sterile water alone
  • A sterile syringe for measuring and transferring diluent
  • Alcohol prep swabs and a clean, aseptic working surface

You can add bacteriostatic water and syringes to any order.

Step by step

  1. Equalise temperature. Let both the vial and the diluent reach room temperature before you begin.
  2. Swab the stoppers. Wipe the rubber stopper of both the peptide vial and the water vial with an alcohol swab.
  3. Add the water slowly. Draw your chosen volume of bacteriostatic water and inject it down the inside wall of the vial, not directly onto the powder. A gentle stream protects the delicate peptide structure.
  4. Do not shake. Swirl the vial gently, or simply leave it to stand. The powder will dissolve on its own within a minute or two. Shaking introduces shear forces and foaming that can damage peptides.
  5. Inspect. A properly reconstituted solution should be clear and free of particulates. Cloudiness or floating material is a sign of a problem.
  6. Record the concentration. Note how much peptide and how much water you used, so you know the resulting concentration for your records.

Working out the concentration

Concentration is simply the mass of peptide divided by the volume of diluent. For example, a 10 mg vial reconstituted with 2 ml of water gives a solution of 5 mg/ml.

Storage after reconstitution

Once in solution, keep the vial refrigerated and sealed between uses, and use aseptic technique each time you draw from it. The benzyl alcohol in bacteriostatic water suppresses microbial growth, but it is not a substitute for careful handling. See our storage and handling guide for detail.
